Project Title: Identification of TDP-43 Low-Complexity Domain Peptides That Promote Aggregation

Summary: Our project investigates the aggregation of TDP-43, a key protein associated with neurodegenerative diseases such as Amyotrophic Lateral Sclerosis (ALS). By combining in vitro assays with recombinant proteins and peptides, we study the molecular mechanisms that drive TDP-43 aggregation. To replicate these processes in cell models, we are developing a stimuli-responsive system to deliver and release medically relevant TDP-43 peptide sequences into cells using a glutathione- and reactive oxygen species (ROS)-cleavable mechanism. This approach allows us to trigger targeted intracellular aggregation and investigate the downstream functional consequences of TDP-43 sequestration.
