This is true in the cytoplasm as well as in the nucleus. An example of the former would be RNA-binding proteins forming stress granules and of the latter the nucleolus. DESCRIPTION:Talk announcement for Prof. Dr. Jörg Höhfeld. \n\nMaintaining the proteome under mechanical stress – from mechanisms to pathology \nBy Prof. Dr. Jörg Höhfeld \nUniversität Bonn · Institut für Zellbiologie \nAbstract from Prof. Höhfeld: \nOrganismal survival depends on the ability to maintain functional protein networks even under conditions that threaten protein integrity. Cells in multicellular organisms are constantly exposed to stress resulting from mechanical forces. In this situation\, protein homeostasis relies on a chaperone machinery that is organized by the co-chaperone BAG3. The BAG3-machinery recognizes force-unfolded cytoskeleton proteins and mediates their disposal through chaperone-assisted selective autophagy (CASA). In addition\, BAG3 interacts with components of the Hippo and mTOR signalling pathways to stimulate transcription and translation. This is important for compensating degradation during cytoskeleton maintenance. In agreement with its essential role under mechanical stress\, BAG3 deficiency causes severe muscle weakness in children and contributes to heart failure\, a leading cause of mortality in industrialized nations. Here\, we discuss the mode of action of the BAG3-machinery and its force-dependent regulation and pathophysiological relevance.

DESCRIPTION:Talk announcement for Prof. Dr. Ralf Metzler. \n\n“Long-ranged correlated processes in disordered systems” \nBy Prof. Dr. Ralf Metzler \nUniversität Potsdam \nAbstract from Prof. Metzler: \nStochastic processes with long-ranged\, power-law correlations naturally emerge in systems such as Rouse polymers or single-file systems\, when all degrees of freedom are integrated out\, apart from the (tracer) particle of interest. In non-equilibrium situations\, the resulting overdamped dynamics corresponds to Mandelbrot’s fractional Brownian motion (FBM). \nThis talk will address some peculiar behaviours of FBM when subject to confinement in box potentials as well as sub- or superharmonic external potentials. Specifically\, it will be argued why superdiffusive FBM may be a good (polymer) model for growing serotonergic fibres in the mammalian brain. \nIn disordered environments it will be shown how non-Gaussian displacement density functions emerge and subtly depend on how the dynamics is introduced. This is achieved by stochastic-diffusivity models connected with long-range dependent noise. Moreover\, it will be discussed how time-dependent critical exponents can be introduced in this formalism. \nFinally\, amoeba systems will be introduced in which the displacement density function appears to converge to a non-Gaussian\, Laplace density.

I will first discuss the physical mechanism of ESCRT-III filaments that drive cell division and cell trafficking in archaea and eukaryotes. I will then discuss the physical mechanism behind the dynamic self-organisation of biochemically active FtsZ filaments in the early stages of bacterial division. Throughout\, I will present the comparisons of our models to in vitro and in vivo data. I will finish up with our recent thoughts on the shapes of symbiotic organisms that might have led to the emergence of eukaryotes.

DESCRIPTION:Talk announcement of Abhishek Singharoy \n\n*Biophysics at the Dawn of Exascale Computers – The Age of Molecular Simulation Bots* \nAbstract:\nMolecular modeling of biomolecular assemblies exemplifies a disruptive area holding both promises and contentions. We will start with a brief story of simulating the first ever cell organelle in molecular details to find how nature has chosen survival fitness over efficiency of energy transfer as an evolutionary design . Despite such advances in exascale computing\, biophysical simulation continues to grapple with handling molecular diversity. So\, we will employ deep learning approaches often used in Google searches\, called the inception network\, to marry interaction signatures from alphafold models and proteomics analysis with predictable patient outcomes.  An application will highlight how molecular modeling is used at an industrial scale to de-risk vector-based vaccines for distribution across 194 countries. DESCRIPTION:Talk announcement of Matthias Altmeyer \n\n*Dynamics and Functions of DNA Repair Compartments* \nAbstract: \nGenome stability is constantly challenged by reactive byproducts of\ncellular metabolism and environmental toxins. Dedicated repair pathways\nhave evolved to deal with DNA damage\, and their malfunctioning is\nassociated with human diseases such as cancer and neurodegeneration.\nElucidation of the hierarchy and temporal order in which DNA damagesensing\nproteins as well as signaling and repair factors assemble around\nDNA lesions has painted a complex picture of tightly regulated\nmacromolecular interactions that build specialized nuclear compartments\nto facilitate repair and maintain genome integrity. While many of the\nunderlying interactions\, e.g. between repair factors and damage-induced\nhistone modifications\, can be explained by lock-and-key or induced fit\nbinding models assuming fixed stoichiometries\, structurally less well\ndefined interactions\, such as the highly dynamic multivalent interactions\nimplicated in biomolecular condensate formation by phase separation\,\nalso participate in the formation of multi-protein assemblies at sites of\nDNA damage. DESCRIPTION:Professors Lin Guo and Rohit Pappu will visit for a talk on May 2\, 2023. \n\n*Hidden complexities of condensates formed by simple systems* \nAbstract: \nApplication of the physics of associative macromolecules has led to growing recognition that condensates\, even of relatively simple systems\, form via hierarchies of processes governed by sequence-encoded molecular grammars that go beyond LLPS. The talk will present data and results from experiments and simulations that highlight the importance of phase separation coupled to percolation for systems governed by homotypic interactions\, and the relevance of the coupling between microphase and macrophase separation in systems where homotypic and heterotypic interactions work together. Broader implications for complex multicomponent systems will also be discussed. \nby Professor Rohit Pappu \nWashington University in St. Louis \nDepartment of Biomedical Engineering \n  \n*Reversing Aberrant Phase Transitions of ALS-linked Disease Protein: FUS* \nAbstract: \nLiquid-liquid phase separation (LLPS) mediates the formation of diverse membraneless organelles\, such as stress granules (SGs) in the cytoplasm. Despite clear biological utility\, dysregulated liquid-liquid phase separation can be detrimental. For example\, phase-separated stress granules enrich RNA-binding proteins (RBPs)\, such as TDP-43 and FUS\, which have an intrinsic tendency to form fibrils that are closely tied to fatal neurodegenerative disorders\, including amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Therefore\, it is important to understand how cells regulate the liquid-liquid phase separation of biomolecular condensates\, which includes maintaining beneficial phases and simultaneously preventing pathological phase separation and fibrillization. As RNA-binding proteins localize mainly in the nucleus\, the phase separation of FUS and TDP-43 can be regulated by both nuclear import receptors and short RNAs. For example\, we found that phase separation of FUS can be regulated by Karyopherinβ2 (Kapβ2)\, which recognizes the proline/tyrosine–nuclear localization signal (PY-NLS) at the C-terminus of FUS protein. On the other hand\, phase separation of TDP-43 can be regulated by the Importina/Karyopherinb1 complex\, which binds to the canonical NLS of TDP-43. In vivo\, Kapβ2 prevents FUS accumulating in stress granules\, restores nuclear FUS localization and function. By preventing and reversing aberrant FUS phase transition and cytoplasmic localization\, Kapβ2 is able to rescue neurodegeneration caused by disease-linked FUS. SUMMARY:Picture a Scientist - an interactive workshop to raise awareness for diversity
DESCRIPTION:Together with the SFB 1361 and GenEvo we are hosting an afternoon workshop to focus on empowerment and to raise awareness for diversity and against discrimination in order to promote a gender- and diversity-sensitive work and research culture. \nScience careers are about the science\, and they’re also about the people – including how people think about\, and treat\, each other. We all want a science system that rewards good ideas and good research and which treats everyone fairly. We also as a group have experienced or suspected (or maybe remained oblivious to) the reality that science systems are not always operating fairly for everyone. And when we lose good ideas and good people to injustices\, we collectively hurt the quality of the science and our own ethics and values. Let’s look together at some of these themes that we inherited from the past and still live with today in order to change the system for the better. \nWe would like to invite all group leaders\, postdocs and PhD students to join us for a productive discussion and lively exchange of views at our first event on Monday\, April 24 2023 4.15-9 pm at IMB\, followed by dinner. What we have planned is: \n\nan interactive workshop led by Prof. Heather Hofmeister\, looking at patterns in our own and others’ experiences in Germany and comparing German academic power structures to those in other countries\, followed by\nthe screening of the documentary feature-length film from 2020 called “Picture a scientist” about the experiences of underrepresented groups in several fields of science\, and afterwards\,\nwork out conclusions and solutions together – and then we get to the pizza!\n\n  \nAbout the moderator \nHeather Hofmeister is a Professor\, Speaker\, Leader\, Coach\, Writer\, and Researcher. She holds a full Professorship for Sociology of Work at the Goethe University\, Frankfurt and is co-director of the Center for Leadership and Behavior in Organizations (CLBO). She researches and speaks internationally on topics in work\, gender\, diversity\, leadership\, careers\, and science\, using a life course and international comparative perspective. \nHofmeister served three years as Vice-Rector at RWTH Aachen University as the first woman in the Rectorate from age 35 and held the first professorship of Sociology with the specialty in Gender and Life Course Research there. In Frankfurt\, she served as Chair of the Department of Sociology\, Chair of the Board of Examiners\, and Dean of Studies at the Goethe University Faculty of Social Sciences. She has led and currently leads large interdisciplinary research projects as PI\, serves on the Board of Trustees of the Technical University of Munich (TUM)\, and served on the Board of Trustees at the University of Bamberg. Hofmeister is a trained coach\, and she facilitates groups and classes of up to 800. And why is it often still “the male scientist” who wins prestige in our imagination? In PICTURE A SCIENTIST\, a female biologist\, a female chemist and a female geologist take on these questions and lead the audience on a journey through the experiences of their academic careers. \nUsing their biographies as examples\, the film deals with a variety of problems that are also relevant in the German university system. These include sexualised harassment in everyday university and research life\, exclusion mechanisms and unequal treatment\, multiple discrimination and the role of leaders and institutions. The film shows the achievements of the past decades\, but also makes visible the resistance women have to face at every level of the academic world.
