DESCRIPTION:Isolating and characterizing proteins for in vitro condensate formation might be problematic! \nHave you ever worked with isolated proteins? Most work in our group involves the isolation of (typically recombinantly produced) proteins plus their biophysical characterization. During this work\, we encountered several problems associated with the insolation of tagged proteins and/or with their biophysical characterization. We have therefore decided to discuss some problems that typically occur but are often ignored “hands on”. We will focus on the isolation of tagged proteins plus their characterization via CD spectroscopy and/or dynamic light scattering. We will use examples from our own research with a condensate-forming protein to highlight some problems and pitfalls.

DESCRIPTION:Talk announcement for Prof. Dr. Jörg Höhfeld. \n\nMaintaining the proteome under mechanical stress – from mechanisms to pathology \nBy Prof. Dr. Jörg Höhfeld \nUniversität Bonn · Institut für Zellbiologie \nAbstract from Prof. Höhfeld: \nOrganismal survival depends on the ability to maintain functional protein networks even under conditions that threaten protein integrity. Cells in multicellular organisms are constantly exposed to stress resulting from mechanical forces. In this situation\, protein homeostasis relies on a chaperone machinery that is organized by the co-chaperone BAG3. The BAG3-machinery recognizes force-unfolded cytoskeleton proteins and mediates their disposal through chaperone-assisted selective autophagy (CASA). In addition\, BAG3 interacts with components of the Hippo and mTOR signalling pathways to stimulate transcription and translation. This is important for compensating degradation during cytoskeleton maintenance. In agreement with its essential role under mechanical stress\, BAG3 deficiency causes severe muscle weakness in children and contributes to heart failure\, a leading cause of mortality in industrialized nations. Here\, we discuss the mode of action of the BAG3-machinery and its force-dependent regulation and pathophysiological relevance.

DESCRIPTION:Talk announcement for Prof. Dr. Ralf Metzler. \n\n“Long-ranged correlated processes in disordered systems” \nBy Prof. Dr. Ralf Metzler \nUniversität Potsdam \nAbstract from Prof. Metzler: \nStochastic processes with long-ranged\, power-law correlations naturally emerge in systems such as Rouse polymers or single-file systems\, when all degrees of freedom are integrated out\, apart from the (tracer) particle of interest. In non-equilibrium situations\, the resulting overdamped dynamics corresponds to Mandelbrot’s fractional Brownian motion (FBM). \nThis talk will address some peculiar behaviours of FBM when subject to confinement in box potentials as well as sub- or superharmonic external potentials. Specifically\, it will be argued why superdiffusive FBM may be a good (polymer) model for growing serotonergic fibres in the mammalian brain. \nIn disordered environments it will be shown how non-Gaussian displacement density functions emerge and subtly depend on how the dynamics is introduced. This is achieved by stochastic-diffusivity models connected with long-range dependent noise. Moreover\, it will be discussed how time-dependent critical exponents can be introduced in this formalism. \nFinally\, amoeba systems will be introduced in which the displacement density function appears to converge to a non-Gaussian\, Laplace density.

I will first discuss the physical mechanism of ESCRT-III filaments that drive cell division and cell trafficking in archaea and eukaryotes. I will then discuss the physical mechanism behind the dynamic self-organisation of biochemically active FtsZ filaments in the early stages of bacterial division. Throughout\, I will present the comparisons of our models to in vitro and in vivo data. I will finish up with our recent thoughts on the shapes of symbiotic organisms that might have led to the emergence of eukaryotes.

This is a joint event organised by SFB 1361\, SFB 1551\, GenEvo\, and 4R-RTG. \n  \nThere will be an interesting talk given by a 2023 Alexander von Humboldt Professorship recipient\, Prof. Hanna Kokko. She is a theoretical evolutionary biologist and is currently based in JGU carrying out research to better understand the interaction between evolutionary and ecological processes. \n  \nThe event is open to all members of the SFB 1361\, SFB 1551\, GenEvo\, and 4R-RTG.
